BrdU 细胞增殖检测试剂盒使用抗 BrdU 抗体检测细胞增殖过程中掺入细胞 DNA 的 5-溴-2'-脱氧尿嘧啶 （BrdU）。当使用含有 BrdU 的标记培养基培养细胞时，这种嘧啶类似物代替胸苷掺入增殖细胞新合成的 DNA 中。去除标记培养基后，固定细胞，并使用我们的固定/变性溶液使 DNA 变性。然后加入 BrdU 小鼠单克隆抗体以检测掺入的 BrdU（DNA 变性对于提高掺入的 BrdU 对检测抗体的可及性是必要的）。然后使用抗小鼠 IgG、HRP 连接的抗体来识别结合的检测抗体。添加 HRP 底物 TMB 以显色。显色的吸光度大小与掺入细胞中的 BrdU 的数量成正比，这是细胞增殖的直接指标。
